Cloning and Gene Synthesis:
1) Cloning of PCR products by restriction enzyme digestion and ligation, TOPO (TA) cloning, In-fusion cloning, Gateway Cloning and Ligation Independent Cloning (LIC).
2) Cloning of your genes into baculovirus, lentiviral, adenoviral and adeno-associated virus vectors. Custom production of high titer integrative or non-integrative lentivirus (108~109 TU/ml), adenovirus (>1012 PFU/ml) and adeno-associated virus (>1013 GC/ml).
3) Gene synthesis (100 bp to 5 kb) and clone synthetic gene into a vector of your choice.
5) Construction of fusion proteins:
* Common Expression and Purification Tags: Poly-histidine Tag (6XHis, 8XHis and 9XHis), Maltose binding protein (MBP), Glutathione-S-transferase (GST), Chitin binding protein (CBP), Thioredoxin, NusA.
* Tandem Affinity Purification Tags: 6XHis.GST, 6XHis.MBP.
* Monomeric Fluorescent Proteins: Fusion of your gene to the N- or C-terminus of EGFP, BFP, CPF, YFP, Red Fluorescent Proteins (RFP), infrared fluorescent protein (IFP), Infrared red fluorescent protein (IRFP), large Stokes shifts RFPs.
* Luciferase and secreted luciferase alone or fused to monomeric fluorescent proteins using self-cleaving 2A peptides (F2A, T2A, E2A, and P2A).
* Detection Tags: c-myc, HA, Flag, 3XFlag, Halo, Strep, T7, S-Tag, Streptavidin-Binding Peptide (SBP), V5 Epitope, Xpress Epitope.
* Color Tags: Soluble Red Tag, cytochrome b5, 13 kDa; Yellow Tag: flavin mononucleotide (FMN) domain of human cytochrome P450 reductase, 21-kDa.
* Secretion Signal Peptides: for protein expression in yeast, insect cells, and 293/CHO cells.
5) Site-directed mutagenesis and multi site-directed mutagenesis.
6) Preparation of endotoxin-free plasmid DNA.
