The self-amplifying NLS-Cre mRNA (SA-NLS-Cre) encodes four
nonstructural proteins (NSP1 to NSP4) derived from a positive-stranded
alphavirus venezuelan equine encephalitis virus (VEEV), and human codon-optimized Cre
recombinase fused to nuclear localization sequence. Upon entering the cytoplasm, the mRNA is
translated to produce replicase complex, which amplifies the original
NLS-Cre mRNA template to produce numerous copies of itself (mRNA
replication), thus driving robust NLS-Cre protein expression. It can be used
for loxP recombination in cell lines and primary
cells
