Background |
Mesothelin gene encodes a
71-kDa precursor protein that is processed to a 40-kDa protein termed
mesothelin, which is a glycosylphosphatidylinositol (GPI)-anchored
glycoprotein present on the cell surface. Mesothelin is a
differentiation antigen that is present on a restricted set of normal
adult tissues such as the mesothelium. In contrast, it is overexpressed
in a variety of cancers including ovarian cancers, non-small cell lung
cancer (NSCLC), colorectal cancer (CRC) and mesothelioma. Its limited
distribution on normal tissues, relatively specific expression at high
levels in tumors and important role in tumor development and prognosis,
make mesothelin a potential target for tumor immunotherapy. |

Flow cytometry assay using R47 monoclonal antibody.
A431 is mesothelin negative cells. A431 (mesothelin++) is derived from
A431 that was forcefully over-expressing mesothelin. One million cells
were incubated with 1 µg antibody in 1 ml PBS buffer containing 5% BSA.
Antibody binding was detected with Cy5-conjugated goat-anti-rabbit
antibody. Blue line, isotype control antibody staining; Red line, R47
staining.
