Description Our functional membrane protein production system is based on state-of-the-art virus-like particle (VLP) technology developed by Excellgen scientists. This platform enables the expression of functional membrane proteins (e.g., GPCRs, receptor kinases, ion channels, viral envelopes) in their native context for downstream assays and screening.
Workflow & Timeline
  1. Cloning and Subcloning (2 weeks):
    • PCR amplification using high-fidelity DNA polymerase
    • Cloning PCR product into an expression vector
    • Restriction digestion analysis and sequencing to confirm correct sequence and frame
    • Preparation of plasmid DNA
  2. Expression Optimization (2 weeks):
    • Transfection of CHO or HEK293 cells with plasmid DNA
    • Assessment of protein expression level by SDS-PAGE, dot-blot, or Western blot analysis
  3. Large Scale Expression and Purification (2 weeks):
    • Transfect 500 ml CHO or HEK293 cells with plasmid DNA (or use baculovirus-Sf9 system as needed)
    • Culture cells using serum-free medium
    • Purify protein using AKTA FPLC systems
    • Dialysis and concentration
    • SDS-PAGE or Western blot analysis of the purified protein
    • Determination of protein concentration
    • Delivery of purified, ready-to-use protein
