Background |
Signal regulatory protein alpha (SIRPα, CD172a) is the primary receptor for CD47, which delivers a "don't eat me" signal to phagocytic cells and is frequently overexpressed on solid tumor cells. SIRPα is predominantly expressed on macrophages and dendritic cells, acting as an immune checkpoint inhibitor for these cell types. The interaction between tumor cell-expressed CD47 and SIRPα on macrophages enables immune evasion by tumor cells. Blockade of the SIRPα-CD47 axis with monoclonal antibodies restores macrophage-mediated phagocytosis of tumor cells, making SIRPα an attractive target for cancer immunotherapy.

Quality Control |
Flow cytometry analysis using M181 monoclonal antibody. A431 cells (SIRPα-negative) and A431 (SIRPα++), which overexpress SIRPα, were incubated with 5 µg antibody in 1 mL PBS containing 5% BSA. Antibody binding was detected with Cy5-conjugated goat anti-rabbit antibody. Shaded area: isotype control; red line: M181 staining.
