Overview |
The D7324 tag (APTKAKRRVVQREKR) is derived from the conserved C-terminus of HIV-1 gp120, corresponding to amino acids 497–511 of the BH10 strain (Ratner L. et al., Nature, 313, 277-284, 1985). Initially, this epitope was used for capturing HIV-1 envelope proteins with a goat polyclonal antibody (D7324) in capture ELISA assays. Subsequently, the D7324 tag has been widely adopted as an epitope tag for recombinant soluble membrane proteins—including those from HIV-1, SIV, Dengue, Zika, and Lassa virus—for capture ELISA in antibody response detection and monoclonal antibody screening. Many recombinant viral envelope proteins are engineered to express a C-terminal D7324 tag, which does not interfere with protein bioactivity. The 1E2 monoclonal antibody specifically captures C-terminal D7324-tagged fusion proteins as well as native Env from lysates of HIV-1 virions or infected cells.

50 µL of 4 µg/mL 1E2 mAb in PBS was coated on an ELISA plate overnight at 4 °C. D7324-tagged protein supernatant from transiently transfected HEK293F cells was captured and detected using a protein-specific detection antibody.
